Study Summary

To determine the maximum tolerated dose (MTD) and the recommended dose (RD) of PM01183 in combination with irinotecan in patients with selected advanced solid tumors

To analyze the safety profile, along with the handling of lurbinectedin and irinotecan in the study population

To obtain preliminary information on the antitumoral activity of PM01183 in combination with irinotecan

To evaluate the pharmacogenomics with tumor samples from patients exposed to both compounds in order to assess potential markers to both response and/or resistance

To evaluate escalating doses of LUR on Day (D) 1 plus a fixed dose of IRI 75mg/m2 on D1 and D8 every 3 weeks (q3w) in pts with advanced solid tumors, enrolled following a standard 3+3 dose escalation design

To evaluate escalating doses of LUR on Day (D) 1 plus a fixed dose of IRI 75 mg/m2 on D1 and D8 every 3 weeks (q3w) in pts with advanced solid tumors (+/- G-CSF, if dose-limiting toxicities [DLTs] were neutropenia)

Phase I: escalating doses of LUR on day 1 plus a fixed dose of IRI 75 mg/m2 on day 1 and day 8 every 3 weeks (q3w) in patients (pts) with advanced solid tumors, following a standard 3+3 dose-escalation design

  • Phase Il expansion at the recommended dose (RD) to further explore efficacy and safety in indications where antitumor activity or a signal of activity was observed during phase I dose escalation
